1. General job accountability:

• Perform total RNA extraction and RT-PCR:

  • Mouse single B cell suspension preparation;

  • Extract high quality total RNA by Trizol reagent;

  • Conduct first strand cDNA synthesis.

• Perform antibody phage libraries construction:

  • Conduct the first round PCR and gel purification of 1st PCR product and sequence quality analysis;

  • Conduct and optimize the second overlap PCR and sequence quality analysis;

  • Prepare high transformation efficiency electro-competent cells;

  • Be responsible for phage library packing and purification.

• Perform antibody phage libraries panning and screening:

  • Perform QC of antigen/biotinylated antigen;

  • Conduct phage solid, liquid and cell panning;

  • Conduct colony picking and culture and induction;

  • Perform high-throughput ELSIA or mirrorball screening;

  • Re-array positive isolates and sequencing;

  • Perform unique sequence analysis and characterization.

• Coordinate lead isolation characterization:

  • Closely work with biology team and support antibody discovery;

• Keep appropriate experimental records and documentation:

2. Qualification and competency requirements：

A. Education/training background:

• 
  Master with 2+ years, or BS with 5+ years of experience Molecular Biology, Biochemistry, cell biology etc.;

• At least 2 years working experience in pharmaceutical or biotech companies is required.

B. Knowledge/Skills:

• 
  Mastery of molecular biology(e.g. PCR, RT-PCR, molecular cloning etc.).

• Hands-on experience in phage display technology.

• Knowledge of antibody properties, structure and sequences is required.

• Knowledge of molecular biology or protein chemistry or structure biology is plus.

• Experience of therapeutic antibody is plus.

• Excellent data management is plus.

• Excellent presentation skills is plus.

• Experience of antibody repertoire analysis is plus.

• Experience of protein structure modeling is plus.

• Experience of following software / tools is plus:

  • Molecular biology: VectorNTI, SnapGene, CLC Workbench, etc.,

  • Data analysis and process: Origin, GraphPad Prism, FlowJo, Spotfire, etc..

C. Others:
