The lab of Haruki Hasegawa in Biologics Department at Amgen South San Francisco is seeking a postdoctoral fellow to investigate the mechanism of intracellular protein crystallization and LLPS induced by the over-expression of various condensation-prone secretory cargo proteins including mAbs and antibody fragments. The lab’s research interests include the elucidation of unique physicochemical properties embedded in proteins that crystallize intracellularly, in-depth characterization of intracellular protein crystallization and protein phase separation events, detailed understanding of cell morphological changes and signaling events during intracellular protein crystallization events, and biotechnological application of ER-associated protein inclusion bodies and intracellular protein crystals.

The initial research projects of the selected postdoctoral fellow are:

characterizing the roles of post-translational modifications and protein-protein interactions in intra-ER protein crystallization event; elucidating the solubility and condensation propensity of a human IgG that induces LLPS protein droplet in the ER lumen during over-expression; exploring the merits of identified effector genes in enhancing difficult-to-express secretory protein production, to list a few potential projects.

The postdoctoral fellow is expected to develop an independent line of research based on initial projects and to present at scientific conferences and publish in leading journals. The successful candidate will operate in a highly collaborative environment, with opportunities to interact with other postdoctoral fellows, scientists, and drug discovery teams while having access to state-of-the art protein expression technologies and resources at core facilities.

The postdoctoral position is for 3 years with the option to extend for an additional year.

Basic qualifications

Ph.D. at the time of appointment

Preferred qualifications

  • Ph. D. in molecular and cell biology, biochemistry, or related life science field
  • Solid publication record and strong oral and written communication skills
  • Extensive cell imaging experience in immunofluorescent microscopy (EPI, confocal, high content, etc)
  • Proficient skills in flow cytometry, cell sorting, intracellular signaling assays, protein purification, and chromatography
  • Cell phenotype morphometric analysis using algorithms and machine learning
  • Solid knowledge on secretory protein biosynthesis, folding, assembly, trafficking, and secretion
  • Familiarity with various stress response signaling pathways and associated cell-based assay methods
  • Research experience in one or more the following areas:
  • Intracellular protein phase separation, protein crystallization, or protein condensation
  • Cell-based assay development and signaling pathway elucidation
  • Recombinant antibody production and expression optimization
  • Biochemical and biophysical characterization of purified recombinant antibodies
