We are looking for a highly motivated and hardworking mammalian expression specialist to join the ASF Protein Technologies group. This laboratory provides comprehensive molecular cloning, protein expression, purification and analytics support for the Amgen pipeline. Amongst the responsibilities will be producing complex multi-specific antibody-like molecules for bioassays and in vivo studies, protein immunogens for therapeutic antibody discovery, and protein reagents which support clinical programs.
Responsibilities
- Work within a team of mammalian expression staff and interact with colleagues in protein engineering, molecular biology and protein biochemistry groups to deliver diverse protein reagents for research programs
- Using transient and stable expression methods, express a wide variety of protein classes; use analytical methods such as Vi-Cell, SDS-PAGE, Octet
- Use knowledge in cell biology and state-of-art expression systems to enhance team’s technical capabilities
- Drive engineering of CHO and HEK 293 cell lines, including the use of genome editing technologies (e.g. CRISPR/Cas9), to increase success with diverse protein classes
- Independently plan detailed procedures for projects including timelines and methods; analyze and interpret scientific data; provide regular updates about project results; document experiments in lab notebooks and LIMS; multi-task with focus on detail in fast-paced environment
- Collaborate with other Amgen Departments such as PKDM and Therapeutic Areas to advance therapeutic programs
Basic Qualifications
- Doctorate degree
Or
- Master’s degree and 4 years of scientific experience
Or
- Bachelor's degree and 6 years of scientific experience
Preferred Qualifications
- Ph.D. degree with a focus on molecular biology or cell biology with strong protein expression component
- Ph.D. and postdoc from a leading institution or biotechnology company
- Biotech/pharma industry experience with emphasis on recombinant protein expression and production, especially with common mammalian transient and stable platforms as well as production in WAVE reactors or other disposable bioreactor technologies.
- Strong knowledge of cell biology, expression construct design, cell line engineering and expression strategies for broad classes of proteins including mAb, bispecific antibodies, immunogens, and membrane proteins.
- Strong knowledge of expression host cell line engineering methods, including genome editing methods (e.g. CRISPR/Cas9), to enhance recombinant protein production and quality
- A track record of productivity, effective verbal and written communication and interpersonal skills, as well as the ability to work in a fast-paced team environment
- Independent critical thinking and scientific achievement in research, exemplified by publications, conference presentations or generation of intellectual property.
