Amgen is seeking a talented and motivated Scientist with expertise in mass spectrometry and other biophysical characterization and analytical techniques to join our Discovery Attribute Sciences team in South San Francisco. In this vital role you will responsible for the implementation of cutting-edge analytical and biophysical techniques for extensive characterization of reagent and therapeutic protein candidates supporting Amgen’s early phase discovery programs.

This individual will lead the reagent protein characterization group at our San Francisco site through cross functional collaborations with groups in Protein Sciences, Molecular Engineering, and various other Therapeutic Areas.

Key responsibilities include:

  • Design and execute fit for purpose mass spectrometry-based experiments to confirm protein identity and report post translational modifications on a wide variety of proteins including immunogens, target proteins, monoclonal antibody critical reagents, mouse surrogates, Proof-of-concept molecules etc.
  • Perform intact and sub-unit mass analysis, post translational modifications, glycan analysis and disulfide link characterization etc. using LC-MS and LC-MS/MS peptide mapping capabilities to further characterize proteins supporting early discovery programs in a timely manner.
  • Execute native MS experiments to analyze protein architectures, stoichiometries and protein ligand complexes.
  • Train protein biochemistry staff on wet lab procedures and operation of the open access MS and assist with mass spectrometry data interpretation where necessary.
  • Lead various outsourced projects through Science Exchange ranging from antibody de novo sequencing to HDX-MS capability etc. to impact screening stage projects.
  • Perform characterization of proteins using SEC, SECMALS/QELLS, DSF, DLS and Edman degradation.
  • Explore existing and new technologies to aid protein characterization efforts through collaborations with local universities and companies.

Basic Qualifications:

Doctorate degree and 2 years of related experience

Or

Master’s degree and 5 years of related experience

Or

Bachelor’s degree and 7 years of related experience

Preferred Qualifications:

  • Deep understanding of mass spectrometry, protein production, post-translational modifications, glycosylation structures etc.
  • Expertise in protein characterization through use of intact mass analysis, glycan analysis, bottom-up, top-down approaches and other biophysical measurements such as SEC, DSF and SEC MALS.
  • Hands on experience using Agilent TOFs and Thermo Orbitrap mass spectrometers including the tribrid series is preferred.
  • Proficiency in mass spec data processing using different platforms such as Protein Metric software, Mascot, Proteome Discoverer and PEAKS software is highly preferred.
  • Experience in protein expression or purification, gel electrophoresis and western blotting is a plus.
  • Ability to multitask, prioritize and work with global teams in a dynamic environment.
  • Excellent managerial skills required to manage and train scientists based across Europe, Asia-Pacific and India.
  • Excellent communication skills (both written and oral), and a proven track record of academic accomplishments (i.e., peer-reviewed publications and/or patents) .
